Research Program

Program Aims:  To identify and understand of the cellular and molecular mechanisms underlying muscle and adipose tissue growth and differentiation, and their manipulation to improve production efficiency. 

 Project Synopses.

1. Leptin – insulin interaction – Implications for energy expenditure and production efficiency. 

Leptin inhibits insulin-mediated lipogenesis in adipose tissue and promotes utilization of free fatty acids by muscle.  Leptin also attenuates insulin-mediated reduction of free fatty acid oxidation in muscle.  Thus, leptin appears to directly modulate an normal array of insulin actions, particularly those associated with the storage and utilization of lipids and free fatty acids.  This interaction is important in over the longer term.  It is controlling basal metabolism and of long-term energy storage and utilization.  The mechanism of interaction between leptin and insulin is being studied from the molecular through to the whole animal level.  Intracellular mechanisms are being studied using a range of models.  An important aspect of this program is interaction with other scientists throughout the United States and internationally.

 2.  The Insulin-Like Growth Factor Axis in Growth Efficiency.

The IGFs are potent stimulators of muscle growth, and are essential for normal growth and development.  Treatment with IGF-1 improves protein acretion by inhibiting breakdown.  This has major significance for manipulation of muscle growth and fat deposition.  The implication is that domestic animals could be managed to be more metabolically efficient, producing more muscle tissue for an equivalent food intake.  There are also extensive potential benefits of these findings in clinical treatment of obesity, muscular diseases, catabolic states such as diabetes, cancer cachexia, and peri-operative stress in humans.  Current investigations focus upon the modulation of IGF actions by IGF-binding proteins.  National and international collaboration is also a feature of this program.


Polyacrylamide gel electrophoresis (PAGE).

PAGE  has proved to be a powerful method used as a component during a protein purification regimen or when characterizing purified proteins. The procedure involves placing a protein into an electric field. Due to the charged nature of the protein, it will migrate towards the electrode of the opposite charge.
